I don't think you'd have to see it from the beginning. If you wanted to start watching it and you did have questions I could answer them for you. They don't follow a tight story line...it's more about getting to know each character. Here are the characters in a nutshell...
Michael Scott - Idiot boss who thinks he's cool and that he's buddies with all of the employess, but he's a big dork. He says inappropriate things all the time which are just SO WRONG but that's what makes it hilarious. He's dating Jan from corporate who is his his boss.
Dwight Schrute - Kiss ass to the boss. Drives a trans am and loves 80's metal...lives on a beet farm. He's a wannabe cop and he's always trying to find ways to work himself into authority positions. He hates Jim because Jim is always fucking with him and Dwight has a secret office romance with Angela from accounting.
Jim - seems to be the only "normal" character besides Pam. He is entertained by the idiots he works with and is constantly finding innovative ways to fuck with Dwight's head. He was secretly in love with Pam the secretary (who was engaged to Roy who works in the warehouse) and confessed to her at the end of last season. She turned him down and he requested a transfer from Pennsylvania to Connecticut. The Connecticut office ended up getting absorbed into the Pennsylvania (Scranton) office so Jim came back and got a promotion. More new characters came onto the show because of this, but now most of them have quit because Michael the boss is an asshole.
Pam - Secretary who is also normal. She and Jim pull pranks on Dwight. She is artistically talented enough to go to art school and make something out of her life but she keeps her job as a secretary (probably to be around Jim). She secretly likes Jim but she couldn't do anything about it when he confessed to her because she was engaged. Now Jim is dating Karen (who was transferred from the Connecticut office) and Pam is suppressing her feelings for him because she is too nice to try to ruin Jim's relationship with Karen.
Angela - works in accounting...she is a total bitch. She's very judgemental, extremely conservative, hardcore Christian, head of the party planning committee, and has a secret romance with Dwight.
Ryan (the temp) - Ryan started out as the temp but he got Jim's old job when Jim was transferred to Connecticut. He would rather be anywhere else than at his job. He's dating Kelly the Indian girl. He secretly feels that he is better than everyone else who works there, he feels that most of the people he works with need to get a life. His facial expressions are hilarious.
Kelly - the most annoying character on the show. She's loud, obnoxious, shallow, and very high maintenance. She's all about drama. She's going out with Ryan and she wants to marry him but he doesn't want to marry her.
Andy - came from the Connecticut office. He's also a kiss ass and wants to be Michael's #2 guy so he's big competition for Dwight. He and Dwight hate each other's guts. Andy is more annoying than Dwight. He hasn't been on the last couple of episodes so I'm not sure what happened to him.
Those are basically the main characters, there are a few side characters that they haven't developed as much like Oscar the homosexual Mexican, Meredith the drunk lush, Stanley the stereotypical black guy who sold out to work for "the man", Phyllis is quiet and kind of blends in, Kevin who is a 40-something immature perv, Creed is an ex-roadie and kleptomaniac, Toby from HR who is hated by Michael because Toby is always having to keep Michael's behavior in check.
Sorry, that's a lot of info but if you know the characters the show would be funnier faster if you're just getting into it. Mostly the show is funny because of the interaction between the characters. There is a lot of subtle humor. The characters all have a relationship with the camera and will look directly into the because the premise of the show is that it's being filmed as a documentary is being made about the office.
